چکیده

Sphingolipids are a diverse group of lipids with important roles in beta-cell biology regulating insulin folding and controlling apoptosis. Sphingolipid biosynthesis begins with the condensation of L-serine and palmitoyl-CoA. Here we tested the effect of L-serine supplementation on autoimmune diabetes development and blood glucose homeostasis in female NOD mice. We found that continuous supplementation of L-serine reduces diabetes incidence and insulitis score. In addition, L-serine treated mice had an improved glucose tolerance test, reduced HOMA-IR, and reduced blood glucose levels. L-serine led to a small reduction in body weight accompanied by reduced food and water intake. L-serine had no effect on pancreatic sphingolipids as measured by mass spectrometry. The data thus suggests that L-serine could be used as a therapeutic supplement in the treatment of Type 1 Diabetes and to improve blood glucose homeostasis.

Oral magnesium supplementation in type II diabetic patients

Background : Magnesium is the second most abundant intracellular cation. It plays an important role in insulin homeostasis and glucose metabolism through multiple enzymatic reactions. With increasing data on magnesium deficiency in diabetic patients and epidemiological studies demonstrating magnesium deficiency as a risk factor for diabetes, it is logical to search for its possible beneficial...

Treatment effect of GABA on improve type one diabetes in NOD mice

Introduction: Gama amino butyric acid (GABA) is the major inhibitory neurotransmitter in the mammalian nervous system. The concentration of GABA and the number of GABA cell secretion decrease in diabetic patient and experimental diabetes model. The reported effects of GABA activation on insulin secretion from beta cells have been controversial. In this study we investigated if GABA administr...

Preventive and therapeutic potential of p38 alpha-selective mitogen-activated protein kinase inhibitor in nonobese diabetic mice with type 1 diabetes.

Mitogen-activated protein kinases (MAPKs) and heat shock proteins (HSPs) are ubiquitous proteins that function within T cells in both normal and stress-related pathophysiological states, including type 1 diabetes. The nonobese diabetic (NOD) mouse spontaneously develops T cell-mediated autoimmune pancreatic beta cell destruction that is similar to type 1 diabetes in humans. Because p38 MAPKs ha...

Zinc Supplementation Improves Glucose Homeostasis in High Fat-Fed Mice by Enhancing Pancreatic β-Cell Function

Zinc is an essential component of the insulin granule and it possibly modulates insulin secretion and signaling. Since insulin resistance is a hallmark in the development of type 2 diabetes mellitus, this study aimed at investigating if zinc supplementation is able to improve glucose tolerance and β-cell function in a model of insulin resistance. Male C57BL/6 mice were distributed in four group...

Inhibition of diabetes in non-obese diabetic mice by nicotinamide treatment for 5 weeks at the early age.

To know the effects of nicotinamide (NCT) treatment for 5 weeks at the early age on insulitis and development of diabetes in non-obese diabetic (NOD) mice, this experiment was performed. Ten ICR (Institute of Cancer Research) and 15 female NOD mice at 4 weeks of age were used. Mice were assigned to ICR and NOD groups, and NOD mice were randomly divided to control and NCT-treated groups. NCT was...
